my next idea is to re-install .. so you are progessing just fine in troubleshooting ..

after you toss prefs and restart .. check your playback engine .. to make sure it came up correctly ..

These interfaces dont require drivers, or so i think. I went on their site, there was nothing, Ive also heard that in the past. Im at my wits end with this it had to be that OS update im telling you.

10.12.5 is latest Sierra ... i was part of the Beta for Sierra last summer ... and there really is nothing that should cause this ..

which Scarlett ?? you did not mention or i missed it .. as there are Drivers for these .. for example i picked Scarlett USB

Really? alright then it must not have been that update, hmmm. oh man this is getting crazy i was paid to record some voice overs due by tomorrow. Its a Focusrite Scarlett solo 2nd gen.

ok, i see no MAc drivers ... only this on gettting started ..

just want to make sure that in Audio / Midi Setup (audio window) that this is where you can see your Scarlett !!! is this correct ..

below is me just finding things that might help .. but you may have already tried all this ...

Mac OS only:
Your OS should automatically switch the computer’s default audio inputs & outputs to the Scarlett Solo. To verify this, go to System Preferences > Sound, and ensure that the input and output are set to Scarlett Solo. For more detailed setup options on a Mac, open Applications > Utilities > Audio MIDI Setup.

Mac Users
1. Connect your Scarlett to your Mac using the included USB cable
2. Go to "System Preferences" then "Sound" and select your Scarlett Eg. "Scarlett 2i2" as the input and output device
3. You are now ready to use your Scarlett audio interface
(Please Note: You may need to select the Scarlett as your interface within your music software preferences)

Mac OS
Apple Macintosh with a USB 2.0 or 3.0-compliant USB port and an Internet connection* OS: Mac OS X 10.10 (Yosemite) or OS X 10.11 (El Capitan)

There have been some issues with Scarlett's and Sierra. See this thread: http://duc.avid.com/showthread.php?t=387434&page=2. About halfway down, some are reposting responses from Focusrite with things to try. Not sure if any of this will help, but worth a try.

ALSO: did you make a backup of your system before you upgraded your OS ???
