Quick fix for a instrument track that won't play
Duplicate a instrument track that is not working. The new track will work.
Yes, I know about the problem and always use aux tracks but this was a outside session. If anyone can confirm this perhaps it will help Digi fix the problem.

If you show instrument in the Mix Window, is your MIDI channel set to '1' ? I saw this (again) the other day. Switching it to '1' fixed it for me.

Yeah, dup'n or making it inactive & active again will often move the MIDI channel to the next available so as Sean said, double check that it hasn't changed...
