Use the DI input as you are taking a line out feed from the channel strip. If you are recording Vocals use a small compression of something like a 3:1 with a soft knee, accoustic guitar shouldn't really need it when recording, but it really depends on the mic you use and where you place it.
When mixing its your baby, it all depends the sound you are after, there are no hard and fast rules, just play until you get the sound you are after.
