I'm on Mac and am not familiar with Midisport
Since you are hearing the EZ drum sounds from triggering on the GUI then I would think it is not midi routing
#1 are you getting signal into instrument to the track (showing in the instrument track meters) from your keyboard ? but simply not getting signal and audio out to the main outs ? If so you might try either a different session or trashing you PT preferences.
If you are not getting any signal into the track from your keyboard then it could be the Keyboard drivers maybe? or a bad USB connection ....

To clarify:: The VI is on an instrument track Yes ?
So in my system this is what I show
Setup > MIDI > MIDI Input Devices > MIDI Input Enable ---all 4, MPK249 (my keyboard ) boxes are checked
PT Preferences >MIDI > Default Thru Instrument > "Follows first selected MIDI track" _____ then I just select/highlight the Instrument track of the VI I want the sound from,(the VI -1 track below) and it works
