I have Back and Play mapped to the non-shift layer RTZ button on the Dock, and that's working fine. I have Play Edit Selection Start mapped as a key command to the shift layer of the RTZ button. The keyboard shortcut is cmd-opt-left arrow. This worked previously in 2022.9. I had backed up my custom soft keys and have tried restoring my back up as well as deleting and recreating the custom shortcut, but it still doesn't work. It works fine using the same key commands on the keyboard.

Thanks to Nathaniel Reichman's suggestion on the FB group, this is fixed. The solution was to remove EuControl and MC_Client from Security& Privacy->Accessibility, log out (reboot in my case) then add them back in. That worked a treat and all is working for me again!
