Assignable knob is where you can put the mouse pointer on any controllable function and you'll see a little blue box around it and then a knob or the wheel can start controlling it. This goes for things like pan, etc but also plugin parameters (as long as the plugin maker allows it.)
You can even lock/unlock control of that parameter (so you can move the mouse and it doesn't have to stay hoovered over the parameter) to the wheel.

it's being described/discussed how it works on the S4/S6 (I've linked just before he gets into it.) It's pretty close to the same for the Dock.
You can assign what knob becomes the assignable knob (other knobs like the monitor knob could be it.)
I have the jog wheel as mine.
