Mbox2 is not "officially supported" on PT12 http://avid.force.com/pkb/articles/en_US/compatibility/Pro-Tools-12-Approved-Audio-Interfaces-and-Peripherals. But, you may be able to make it work anyway...

Isn't the whole point of the PT "automatic large fixed playback buffer for mixing (2048 samples at 96 kHz - High Latency domain)" - so you don't have to (even can't!) change buffer...
