You would first have to save the instrumental track to your computer.
Create a new session in Pro Tools. You can use a blank session.
From the Pro Tools File menu, select Import -> Audio. This will convert and import the audio file onto a new track in your session.

Now create a new mono audio track and assign the input to the input of the Scarlett where you have the mic connected.

Arm the track for record, and now you will be able to record your vocal along with the music.

You can always use the Web Based Manual for reference.
