Anyone use this configuration? Now I'm on Yosemite with HDX and 12.8.1.
Can I upgrade my maOS to High Sierra? Or is best to upgrade to Sierra?

12 core 2.66 here. HD 2018.1 seems not as stable as 12.8.3 is HS.
Although I'll venture 2018.1 without clients. HS is a completely different drive/file type which also requires a 5,1 Mac Pro firmware upgrade. It also lends itself to SSD's although I've used Firecuda 7200 rpm hybrid drives as well. Just keep in mind this is not your usual OS change. Why the need for Yosemite (10.10.x) change? Maybe start by trying HD 12.8.3 of Pro Tools. This is a solid version.

Not HDX but so far HDN on High Sierra ( 10.13 .2 ) and PT HD 2018.1.0 is working great
On mid 2010- 6 Core Westmere 333 with PCIe SSD as boot drive. (see sig line) Be aware High Sierra uses and installs Apple's new APFS file system and it is not recommended for Hard Disk drives (may work, but read several articles that did not recommend it) So your boot drive (at least) should probably be an SSD Also note 2018 is qualified for HS 10.13.2 And note to DetroitT I am curious because my Mid 2010 Mac 5,1 did not require a firmware upgrade ? Is your machine a Westmere

Yes, my Mac Pro is a Westmere 3,33 Mid 2010 with Yosemite and SSD for boot. My idea is to upgrade to High Sierra and Install PT HD 2018.1.
The question is: I make a backup with time machine and upgrade to High Sierra the current SSD or I buy a new SSD and I made a clean installation of HS?

Then erase your current SSD then install PT Also I remember somewhere it was recommended to install the HD Drivers before installing Pro Tools which is what I do But this time I actually just trashed PT Preferences , installed HS over Sierra and then HD 2018 Drivers and then PTHD 2018 over PTHD 12.8.2 And so far no problems

I had great experience with my clean install of High Sierra, but as soon as I ran a point update to macOS everything went unstable, so the jury is still out there...
