Humm? Aren't the platters the same size in a 3.5" drive no matter the capacity? You won't see any real difference if you partition.

When writing, the computer attempts to write the date in one continuous file. It doesn't jump around unless the drive is very fragmented and doesn't have continuous free space.

One way to de-fragment a file is to copy it to another drive.

A good reason to partition is to put different operating systems on one drive. You would make a partition for each OS. Another is if you want to keep user files separate from the OS. It may be easier to backup just the user files that way. Also, some users like putting a swap file in a different partition.

If the drive is to be used for audio files, no partition is necessary. By default, Mac OS X systems also use a single partition for the entire filesystem, but use a swap file inside the file system rather than a swap partition.
