850 Pro is a SATA III SSD. SATA III SSDs deliver around 500MB/sec , and your Cheesegrater only has SATA II on the motherboards so you will be getting ~250-300 MB/sec unless you install a SATA III PCIe card... but why do SATA anything when PCIe/NVMe based M.2 drives are so very affordable and have way better performance. A good M.2 drive with 4 x PCIe 3 interface (like a Samsung 970) today can deliver reads at around 3.5GB/sec. So more than an order of magnitude faster. Your crappy current HDD,... I won't even embarrass you by saying how slow they are once you factor in a little disk seeking.

Your cheesegrater has PCIe 2 slots so if you want to get the best performance from PCIe 3 based SSD today you need to use a switch based PCIe slot to M.2 adapter card (basically a Syba I/O Crest SI-PEX40129). This stuff has been discussed to death in past threads here, you can search for those.

You are in the stone age if running on HDD... but be careful rushing to change storage, go to NVMe etc. Read and learn first.

I say, get this:
Sonnet Technologies Tempo 6Gb/s SATA PCIe 2.0
It's $100, put your SSD on it, and clone your startup disk to it, and enjoy the new speed and sparkle it gives your mac in the form of boot times, launch times, and such. 500mb/s read/write instead of 100mb/s.

It's very easy and quickly done.

Later you can move on.

The Accelsior S from OWC I use holds one ssd for $50 and also provides approx. 500mb/s read/write. All depends whether one wants to host one or two ssd's in one pcie slot. I'm happy with what I've got and it didn't break the bank nor do I have to worry about heat issues with an nvme type drive.

I wouldn't worry about the so called heat issues. Have been running NVMe for years already and never installed the recommended heat sink.

Having the MacOS SSD connected to a PCIe Slot or the Audio HD?

Both.
OS on fast drive: Boot, app launch, general.
Sessions on fast drive: Sessions load, audio processing, saving, copying, so on.
They could be two separate, or one larger.

Do you use much in the way of samples? If so I'd put that on an ssd in a pcie slot along with the system ssd on a pcie adapter card. Leave sessions where they are now. If the answer is no on samples then why not do both on ssd's on pcie adapters cards?

Where do you see most need for throughput performance? Samples first, audio second, OS last.

Where do you see most need for low latency? OS first, samples second, audio last.

So I would have my OS on any SSD because of low latency (no seek time) but any SSD will do, even "slow" SATA bay because OS does not need so much throughput and how often you boot anyway? (Before the recent 10.15.5 update my uptime was 45 days). The real question is whether your fastest SSD is fast enough for sample based virtual instruments and audio sessions at the same time. If not, most likely your OS and audio sessions can share same SSD (because OS needs low latency and session needs good throughput).
