Sorry if this is already been covered been covered.
Whilst running Media Composer on a a very similar setup to yours (MacPro 5.1, 10.10.5 with dual 3.2 Xeons) here I had been getting both video and or audio dropouts with a very large current project that uses a variety of video media formats.
Are you running the video on a separate drive. If not I would recommend doing that and making that drive an SSD if possible.
FWIW What helped here was fitting a PCIe card (less than $150) with SSD drive mount. SSD performance is stellar. The card has an added bonus of a SATA connection that I used to an external SATA drive. This SATA drive showed nearly 3 times gain in Read Write speeds (4GBs) over it's performance when mounted internally. (BM speed test)

are you? Doesn't sound like it since you have audio dropouts too, but worth checking.

It's been a bug for about 3 years that hasn't been fixed and it's not random. Happens 100% of the time with given settings:

PT is in loop playback
Pre-roll is on
and the video sync offset is not 0.

The video will do a 1-2frame "black wink" right on the edge where the pre-roll ends (where you hit play)

I am not talking about the black wink as its indeed an old bug.
I describe the situation when while screening 120 minutes feature, there might be a 5 sec blackout e.g after 45 minutes and its totally unpredictable on which minute and if it's gonna happen again.
But it's quite frustrating when having a client with 12 other guests sitting in your room.
Happened to me just before yesterday.
