To make use of the Structure 1.0.1 update you have to own a Structure 1.0 license, and have Structure Professional Sampler Workstation 1.0 installed. This update is not usable for Structure LE or Structure Free, the relevant bugfix is already included in the 1.0 versions of these.

What’s Fixed in Structure 1.0.1?
Structure 1.0.1 contains the following fix:

The Structure 1.0.1 update speeds up loading of samples that are distributed across multiple big files. The perceived loading speed especially of large or complex patches gets drastically minimized. This particularly comes in handy when working with Symphonic Orchestra Gold Complete—Structure Edition.
