We justt installed Tiger and get the message "Unable to locate Digidesign Hardware. Make sure your hardware is connected and turned on.", when trying to start Protools. Any ideas?

Maybe you've done this already, but have you power cycled your audio interface? If you have a 192 are the sample rate and sync mode lights lit? I've seen systems where the audio interface doesn't start up properly all of the time and needs a power cycle (wait until the lights are on before moving on). Most likely you are aware of this and you are having some other problem, but just in case I'll mention it.

Thanks Steve. Yes, all six of the 192 I/O's are lit and show 48 sample rate and INT for SYNC MODE. Any other ideas?

Reinstall PT.
Upgrading the OS will remove files that PT needs in the "system" folder.

Have you any other applications opened? It happened to me when I had Sibelius or other applications opened.
