I was getting constant AAE -6101 errors and upping my hardware buffer size with no improvement.
I tried disabling plugins and a host of other things. Switching the playback engine to Pro Tools aggregate seemed to solve the problem but it left me with a useless factory 003 interface.
Looking in the optimisations on the Avid Knowledge Base, I turned off the firewire networking and this solved the problem.

Disable FireWire Networking (FireWire Devices only)
This optimization can help free up bandwidth in the firewire bus and allow Digidesign FireWire devices to work more efficiently.
Open System Preferences.
Click 'Network'.
On the left hand sidebar, click on FireWire/Built-In FireWire/Thunderbolt FireWire.
Click on the drop-down menu for 'Configure IPv4' and choose Off.
Click Apply and restart your computer.
