Load the correct Eleven Rack driver for whatever version of Windows you are using. Then check in your "control panel" under "device manager" and under "Sound, video and game controllers" to make sure that Windows recognizes the Eleven Rack.
If it does, you should be able to select the Eleven Rack for your Skype audio device.
Connecting the Eleven Rack's SPDIF "out" to the Asus soundcard SPDIF "in" will also require the correct driver and is more involved. You would have to set the Eleven Rack's clock to the Asus soundcard. The driver control applet or the 11R itself can be configured to do that.
